ABSTRACT

A 26 year old male patient presented with abdominal distension following ingestion of liquid nitrogen and was diagnosed to have massive pneumoperitoneum both clinically and radiologically. Patient underwent emergency laparotomy in search of perforation, the stomach, Intestines was examined and no hollow viscous perforation was found. All abdominal contents were normal, no tissue necrosis or haemorrhage, no free ?uid or bleeding was found in the abdomen. Two abdominal drains were placed in the lesser sac and the pelvis and abdomen were closed. The patient was stable post operatively and tolerated oral feeds and discharged on 7th post operative day. Our study revealed that despite of having massive pneumoperitoneum, site of perforation cannot be identi?ed

ABSTRACT

RESUMEN Neltuma alba Griseb es una especie emblemática de la Ecorregión Chaqueña, por el valor de su madera y frutos. Los resultados de la presente investigación, dan cuenta de que las semillas de algarrobo blanco pueden ser críoconservadas sin afectar el poder y la energía para germinar. Las semillas fueron conservadas por tres meses a Temperatura ambiente (24° y 30°C, Congelamiento (-18°C) y Nitrógeno líquido (-196°C). Posteriormente, fueron escarificadas mecánicamente y sembradas en tierra negra y aserrín (3:1). Los resultados fueron sometidos a pruebas no paramétricas de Kruskal-Wallis, analizándose poder, energía y vigor germinativo, y no se detectaron diferencias significativas entre tratamientos. Para la regeneración in vitro en medio de cultivo MS suplementados con 3 mg L-1 IBA y 0,05 mg L-1 CIN, se utilizaron segmentos nodales de 2 cm de longitud obtenidos de plántulas provenientes de semillas. Los porcentajes de enraizamiento obtenidos fueron elevados (87-90%). No existiendo diferencias significativas entre los tratamientos ensayados. El almacenamiento de semillas a 24°-30°C resulta ser el más apropiado para alcanzar altos porcentajes de germinación


ABSTRACT Neltuma alba Gris is an emblematic specie of Chaco Ecoregion for its wood and fruits value. This research show that algarrobo blanco's seeds can cryopreserve without affecting germination power and energy. The seeds were kept for three months at 24°-30°C, (-18°C) and (-196°C). Later, they were mechanically scarified and sown in black soil and sawdust (3: 1). The results were subjected to non-parametric Kruskal-Wallis tests, analyzing germination power, energy and vigor. The test did not detect significant differences between treatments. The nodal segments (2 cm long) obtained from plants of seeds were used for in vitro regeneration in MS culture medium supplemented with 3 mg L-1 IBA and 0.05 mg L-1 CIN, The rooting percentages obtained were high (87-90%). There are no significant differences between treatments tested. The storage of seeds at 24°-30°C is the most approppiate to rich high percentages of germination.

ABSTRACT

RESUMEN: El queratoquiste odontogénico (QQO) es una quiste intraóseo poco frecuente que varía entre un 3 a 11% de todos los quistes odontogénicos, su ubicación en el maxilar es rara y la invasión al seno maxilar lo es aún más. El QQO es una patología benigna, localmente agresiva que tiene una alta tasa de recidiva. Se han descrito diversas técnicas quirúrgicas para su tratamiento, las cuales van desde lo más conservador como la enucleación a lo más radical como una resección. El uso de agentes coadyuvantes químicos o cauterizantes han logrado disminuir la tasa de recidiva en conjunto con tratamientos más conservadores, disminuyendo la morbilidad y secuelas asociada a una resección. El objetivo de este trabajo es presentar una serie de casos clínicos de QQO que invaden el seno maxilar, su tratamiento de manera conservadora y una revisión de la literatura comparando los diversos tratamientos y su tasa de recidiva.


ABSTRACT: Odontogenic keratocyst (OC) is a rare intraosseous pathology that varies between 3 % and 11 % of all odontogenic cysts, its location in the maxilla is rare, and invasion of the maxillary sinus is even more so. OC is a benign, locally aggressive pathology that has a high recurrence rate. Various surgical techniques have been described for its treatment, ranging from the most conservative, such as enucleation, to the most radical, such as resection. The use of chemical or cauterizing adjuvant agents has managed to reduce the recurrence rate in conjunction with more conservative treatments, reducing the morbidity and sequelae associated with a resection. The objective of this work is to present a series of clinical cases of OC that invade the maxillary sinus, their treatment being carried out in a conservatively manner, and a review of the literature comparing the various treatments and their recurrence rate.

RESUMEN La crioterapia es el conjunto de procedimientos que utilizan el frío en la terapéutica médica. Emplea diversos sistemas y tiene como resultado la disminución de la temperatura de la piel; produce una destrucción local de tejido de forma eficaz y controlada. El objetivo de este trabajo fue realizar una actualización para exponer los aspectos esenciales sobre formas de empleos, indicaciones, complicaciones y contraindicaciones. Existen varios métodos de aplicación de la crioterapia, que incluyen las técnicas de congelación de spray o aerosol y con aplicadores, el método criosonda, y el uso de termoacoplador. Está indicada en varias entidades, entre las que se encuentran la queratosis seborreica y actínica, lentigos solares, carcinoma basocelular y espinocelular in situ. Las complicaciones más observadas son vesicoampollas, hiperpigmentación e hipopigmentación, y las contraindicaciones comunes son intolerancia al frío, tumores con bordes no delimitados o con pigmentación muy oscura, en localizaciones cerca de los márgenes de los ojos, párpados, mucosas, alas nasales y el conducto auditivo. El dominio de los métodos de aplicación e indicaciones es indispensable para elegir la conducta adecuada; de esta forma se evitan complicaciones y efectos colaterales (AU).


ABSTRACT Cryotherapy is the whole of procedures that use cold in medical therapy. It uses various systems and results in a decrease in skin temperature, leading to a local destruction of tissue in an effective and controlled way. The objective of this work is to make an update to expose the essential aspects on the ways of use, indications, complications and contraindications. There are several cryotherapy application methods that include spray or spray freezing techniques and applicators, the cryoprobe method, and the thermocoupler use. It is indicated in several entities, and among the most frequent are seborrheic and actinic keratosis, solar lentigo, basal cell and squamous cell carcinomas in situ. The most observed complications are vesical blisters, hyperpigmentation and hypopigmentation, and the most common complications are: cold intolerance, tumors with non-delimited borders or very dark pigmentation, located near the margins of the eyes, on eyelids, mucous membranes, nasal wings, and on the ear canal. The mastery of the signs and application methods are essential to choose the appropriate behavior against the disease: side effects and complications are avoided that way (AU).

ABSTRACT

OBJECTIVE@#To compare the clinical therapeutic effect of fire needling stripping after local anesthesia, simple fire needling and liquid nitrogen cryotherapy on verruca vulgaris.@*METHODS@#A total of 900 patients with verruca vulgaris were randomized into a fire needling stripping group (300 cases, 2 cases dropped off), a fire needling group (300 cases, 4 cases dropped off) and a liquid nitrogen cryotherapy group (300 cases, 5 cases dropped off). After local anesthesia of compound lidocaine cream, fire needling therapy was adopted, and the necrotic tissue of verruca was stripped in the fire needling stripping group. Simple fire needling therapy was adopted in the fire needling group, without local anesthesia and stripping. Liquid nitrogen cryotherapy was adopted in the liquid nitrogen cryotherapy group. The treatment was given once a week, and totally 3 weeks were required in the 3 groups. The skin lesion scores of number, area, thickness, color, pruritus, isomorphism and the level of T lymphocyte (CD@*RESULTS@#Compared before treatment, the skin lesion scores were decreased (@*CONCLUSION@#Fire needling stripping after local anesthesia can effectively treat the verruca vulgaris, improve the skin lesion and immunity, its therapeutic effect is superior to simple fire needling and liquid nitrogen cryotherapy.

ABSTRACT

Dos atendimentos ortopédicos realizados em aves no HCV-UFRGS, 86% são fraturas, sendo aproximadamente 30% delas cominutivas com perda óssea expressiva, justificando a importância da utilização de enxertos em fraturas de aves. O objetivo deste trabalho foi avaliar dois aloenxertos e enxerto sintético de hidroxiapatita em defeito ósseo de galinhas. Utilizaram-se 30 galinhas separadas em três grupos: aloenxerto congelado em ultra-freezer (GUF), aloenxerto congelado em nitrogênio líquido (GNL) e enxerto sintético de hidroxiapatita deficiente em cálcio (GHA). Nos três grupos, os enxertos foram aplicados com placas e parafusos bloqueados de 2mm na ulna direita das aves, avaliando-se a evolução por meio de exames radiográficos até serem completados 90 dias de pós-operatório e o resultado final mediante exame histológico. A média e desvio-padrão relacionando o tempo de consolidação óssea radiográfica foi: GNL 61,67±21,79 dias (90% de consolidação), GUF 47,14±13,50 dias (70% de consolidação) e GHA 70±18,17 dias (60% de consolidação). Houve diferença significativa no tempo de consolidação óssea entre o GUF e o GHA. Histologicamente, os enxertos do GUF foram os que estavam em consolidação mais avançada. Os aloenxertos do GNL foram superiores no preenchimento de falha óssea ulnar de galinhas.(AU)


Of the orthopedic visits performed on birds at HCV-UFRGS, 86% are fractures, and approximately 30% of them are comminuted with expressive bone loss, justifying the importance of the use of grafts in bird fractures. The objective of this work was to test two allografts and a synthetic HADC graft on finishing in Gallus gallus domesticus. 30 laying hens were used, divided in three groups: frozen allograft in ultrafreezer (UFG); frozen allograft in liquid nitrogen (LNG); calcium deficient synthetic hydroxyapatite graft (HAG). The three graft groups were exposed to serial radiographs until the 90 postoperative days, as well as the histological examination at the end of the experiment were: LNG 61.67±21.79 days (90% consolidation), UFG 47.14±13.50 days (70% consolidation) and HAG 70±18.17 days (60% consolidation). There was a significant difference in bone healing time between GUF and GHA. Histologically, GUF grafts were the ones that were in the most advanced consolidation. LNG allografts were superior in filling ulnar bone failure of fowl.(AU)

ABSTRACT: The objective of this study was to determine the ideal moisture degree for the cryopreservation of Cattleya labiata Lindl. and Miltonia regnellii Rchb.f. orchid seeds. Orchid seeds were standardized in relation to the desired moisture degrees (4, 6, 8, 10, 12, and 15%) and were subjected to rapid freezing in liquid nitrogen (-196 °C) for 24 h; then, the seeds were defrosted and analyzed. For both orchid species, the seeds that were immersed in liquid nitrogen with 4% moisture content had the highest viability percentage, averaging 95% and 68% of viable seeds for C. labiata and M. regnellii, respectively. The seeds with 12% and 15% moisture content had no viable seeds. For the fresh mass of seedlings, there was no significant difference between moisture content treatments for either species.


RESUMO: O objetivo deste trabalho foi determinar o grau de umidade ideal para criopreservação das sementes das orquídeas Cattleya labiata Lindl. e Miltonia regnelli Rchb.f.. As sementes das orquídeas foram padronizadas em relação aos graus de umidade desejados (4, 6, 8, 10, 12 e 15%) e submetidas ao congelamento rápido em nitrogênio líquido (-196 ºC) durante 24 horas; em seguida, descongeladas e analisadas. O maior percentual de viabilidade, tanto para as sementes de C. labiata quanto para M. regnelli, ocorreu quando as sementes foram imersas em nitrogênio líquido com 4% de umidade, atingindo 95 e 68% de sementes viáveis, respectivamente, sendo que, as sementes com 12 e 15% de umidade não apresentaram sementes viáveis. Em relação à massa fresca de plântula, não houve diferença entre os tratamentos (grau de umidade)para as espécies avaliadas.

ABSTRACT

In the present study, the performance of the liquid nitrogen frozen and thinned bovine pericardium was studied and compared with the porcine pericardium. The microstructure and mechanical properties of the bovine pericardium were observed and tested by hematoxylin-eosin (HE) staining and tensile test respectively. In all conditions, porcine pericardium was selected as a control group. The results showed that there was little difference in the performance of bovine pericardium after being frozen by liquid nitrogen. The secant modulus and ultimate strength of the thinned bovine pericardium were similar to those of porcine pericardium, however, the elastic modulus was a little higher than porcine pericardium. The study suggested that the performance of the thinned bovine pericardium was similar to those of porcine pericardium. It was easy for the thinned bovine pericardium to obtain a relatively ideal thickness and expected performance, therefore, the thinned bovine pericardium can be used as the materials of transcatheter aortic valve leaflets.

ABSTRACT

We report our experience with a case of stomach perforation after accidental ingestion of liquid nitrogen. A 13-year-old boy ate a snack at an amusement park and began to complain of sudden onset of severe abdominal pain with shortness of breath. It was determined that the snack he had ingested had been cooled with liquid nitrogen. A computed tomography scan of the abdomen and a chest X-ray showed a large volume of pneumoperitoneum. During surgery, a 4-cm perforation of the angularis incisura of the stomach was identified. Primary repair and omentopexy was performed. The patient was discharged without postoperative complications.

Objective Three kinds of pretreatment methods were studied to compare and select the suitable pretreatment methods for Lycii Fructus (LF) reference standards to improve its crushing and packaging efficiency. Methods LF was treated by three pretreatment methods of decompression drying, liquid nitrogen freeze-drying, and freeze-drying. The difference of characteristic values before and after treatment was determined and compared with LF without pretreatment. The determination of characteristic values included the determination of moisture, content, etc. Results The water content of raw material of directly crushed LF was 6.4%, and the water content of the other three pretreated LF was 4.6%, 5.0%, and 5.2%, respectively. The content of polysaccharide and betaine in the raw material of directly crushed LF was 2.5% and 0.98%. The polysaccharide content of the other three pretreated LF was 2.6%, 2.8%, and 2.6%, respectively. The content of betaine was 0.94%, 1.00%, and 0.87% by the other three pretreatment methods. The results showed that there was no significant difference in the quality of LF obtained by the three pretreatment methods. Therefore, from an economic point of view, this paper recommended to use more affordable pretreatment method of decompression drying for LF. In addition, the pretreated raw materials of LF should be crushed and subpackaged as soon as possible, and the crushing and the dividing environment humidity should be strictly controlled at 20% to 30%, in order to improve the efficiency of crushing and dividing. Conclusion Based on the cost consideration, the decompression drying method is the most economical and effective of the three pretreatment methods. Therefore, the decompression drying method is recommended in actual work.

RESUMEN La extracción de ARN de calidad constituye el primer paso para el análisis de la expresión génica. Sin embargo, su obtención no es sencilla debido a la susceptibilidad de esta molécula a la presencia de contaminantes como ARNasas, proteínas y polisacáridos. Adicionalmente, debido a la diversa composición de la pared celular de los hongos se requiere optimizar los procesos de extracción de ARN para organismos específicos. Este estudio evalúo el uso de diferentes metodologías de homogeneización de tejido (nitrógeno líquido y liofilización) y extracción de ARN (Trizol, CTAB y RNeasy mini kit) a partir del hongo nativo ascomiceto Xylaria sp. Se determinó la pureza, concentración e integridad del ARN obtenido por medio de espectrofotometría y electroforesis. Adicionalmente, se diseñaron cebadores de referencia para el gen β-Tubulina a partir del alineamiento de secuencias de este gen obtenidas de diferentes ascomicetes. Estos cebadores fueron utilizados para evaluar si el ARN extraído es amplificable mediante RT-PCR. Se determinó que la homogeneización de tejido por medio de liofilización generó mayores rendimientos de extracción independientemente del protocolo de extracción utilizado; sin embargo, éstos alteraron la integridad del ARN. Se obtuvo un ARN con mayor pureza con el protocolo CTAB y un mayor rendimiento con el RNeasy mini kit. Los resultados indican que el ARN extraído, independientemente de la metodología de homogeneización y extracción utilizada, es amplificable mediante RT-PCR. No obstante, se recomienda homogeneizar el tejido con nitrógeno líquido y extraer con RNeasy mini kit por la brevedad del protocolo de extracción y calidad obtenida.


ABSTRACT Obtaining high quality RNA is the first step for gene expression analysis. However, the low stability of this molecule and high presence of contaminants such as RNases, proteins and polysaccharides may trouble extractions. Fungi cell wall composition is highly diverse; therefore optimizing RNA extraction procedures is necessary when studying specific organisms. In this study, different methods of tissue homogenization (liquid nitrogen and lyophilization) and RNA extraction (Trizol, CTAB and RNeasy mini kit) were assessed with a native ascomycete, Xylaria sp. RNA purity, concentration and integrity were determined by spectrophotometry and electrophoresis. In addition, a set of housekeeping gene primers was designed targeting the β-tubulin gene. The primers were used to determine if the RNA extracted allowed RT-PCR amplification. It was demonstrated that homogenization of tissue by lyophilization allowed higher yields of RNA regardless of the extraction protocol used, however, the RNA integrity was affected. The higher RNA purity was obtained using CTAB and the higher yields using the RNeasy mini kit. The extracted RNA is amplifiable by RT-PCR regardless of the homogenization and extraction methodology used. However, it is recommended to homogenize the tissue with liquid nitrogen and to extract RNA with the RNeasy mini kit due the shortness and efficiency of these protocols.

We discussed the influence of liquid nitrogen cryopreservation to survive capability of Babesia microti standard strain.The whole blood of mice infected with Babesia microti was put in liquid nitrogen to cryopreservation for 1 month,3 months,6 months,9 months,the whole blood was get out respectively and recovery at room temperature,and infected 3 mice respectively,100 μL/ mouse (the first generation after redissolution,the experiment group).In the same time,3 mice were also infected with Babesia microti as the animal conservation control group.When the infection rate was at a high level,the whole blood of the experiment group mice were injected into 3 normal BALB/c mice (the second generation after redissolution),to observe the changes of the Babesia microti form and proliferation situation,and also to observe the infection rate of the first and the second generation after redissolution in different conserving time.Compared with Babesia microti of animal subcultivation,the form of Babesia microti of liquid nitrogen cryopreservation changed a little.Small trophozoites,annular trophozoites,schizont and immature and mature merozoite and other form can also be seen.Compared with Babesia microti of animal subcultivation,the first time to see the worms and the time attaining to the high infection level were 1 to 2 days later,but for the second generation after redissolution,it is the same.There was no significant difference in different conserving time of 1,3,6,9 months.The influence of liquid nitrogen cryopreservation to survive capability and worm form of Babesia microti is a little,so liquid nitrogen cryopreservation can be a better way to conserving Babesia microti.

ABSTRACT

El osteosarcoma es el tumor óseo primario, maligno, más común; se presenta más frecuentemente en la segunda década de la vida. Las metáfisis de los huesos largos que crecen rápidamente como el fémur distal y el húmero proximal son comúnmente los más afectados. Clínicamente es característico el dolor óseo persistente por 3 ­ 6 meses. Radiológicamente, presenta imágenes de márgenes no geográficas, con destrucción (lisis) y diferentes tipos de reacción perióstica, incluyendo el triángulo de Codman. El principal tratamiento incluye la quimioterapia y la cirugía, donde según su estadío, se plantea la conservación del miembro, plastia rotacional o amputación. En éste trabajo presentamos un caso clínico de un paciente adolescente con osteosarcoma de fémur tratado con cirugía preservadora del miembro con la técnica de criocirugía (nitrógeno líquido)(AU)


Osteosarcoma is the most common bone malignant tumour, frequently appearing during the 20s. The metaphyses of long bones which have a rapid growth, e.g. knee and shoulder, are the most commonly affected. A clinical characteristic is the persistent pain in the affected bone which may last from 3 to 6 months. Radiologycally, the lytic areas have non-geographical boundaries and present destruction of the bone tissue, apart from having different types of periosteal reactions, including the Codman Triangle. The treatment consists of chemotherapy and surgery. Chemotherapy is applied before and after surgery. The surgery may be limb salvage surgery or complete amputation of the affected member. This work describes a clinical case of an adolescent suffering from a femur osteosarcoma treated through limb salvage surgery under cryosurgery(AU)

ABSTRACT

La hiperplasia multiepitelial focal (HMF), se caracteriza por la aparición de lesiones múltiples en la mucosa de la cavidad bucal en población pediátrica y se asocia con la presencia del virus papiloma humano. Actualmente no existe un tratamiento de elección, el más empleado es el nitrógeno líquido (NL), pero es doloroso e incómodo para el paciente. Es por eso la necesidad de buscar nuevas alternativas de tratamiento, cuidando de ser menos invasivos, como el ácido glicirricínico (AG). Objetivo: Comparar la eficacia del AG contra el NL en la HMF en pacientes entre los 5 a 13 años de edad, en el HIM. Determinar las características clínico-epidemiológicas y los efectos adversos. Material y métodos: Ensayo clínico controlado aleatorizado. Se dividió en dos grupos. Grupo A tratado con la aplicación de AG cuatro veces al día con dos disparos del atomizador por un mes. Un grupo B tratado con NL una vez al mes por tres meses. Resultados: Se estudiaron 20 pacientes en edad pediátrica, 11 masculinos y 9 femeninos; 12 con familiares afectados. La topografía más frecuente fue el labio inferior. El grupo tratado con AG tuvo una eficacia de 63% en comparación con NL que tuvo una eficacia del 81%. Sólo se observaron efectos adversos sistémicos con NL. Se obtuvo una p de 0.075 (7.5%). Conclusiones: Se comprobó la eficacia del AG, a pesar de que fue menor que con NL. In pediatric population, focal multiepithelial hyperplasia (FMH) is characterized by the onset of multiple lesions in the mucosa of the mouth; it is associated to the human papillomavirus. Presently, no special treatment is preferred, the most widely used is liquid nitrogen (LN) treatment, nevertheless, this is an uncomfortable and painful treatment for the patient. This would prompt us to look for new, less invasive treatment alternatives such as use of glycyrrhizinic acid (GA). Aim: To compare GA efficiency versus LN in FMH in Mexico Children's Hospital patients (HIM) aged 5 to 13 years. To determine clinical epidemiological characteristics as well as adverse effects. Material and methods: The present was a randomized controlled clinical trial. It was divided into two groups: group A was treated with GA applications, four times a day, two atomizer shots, for one month. Group B was treated with LN once a month for three months. Results: 20 pediatric-age patients were studied, out of which 11 were male and 9 female. 12 patients had relatives afflicted with the disease. Most frequent topography was the lower lip. Efficiency of group treated with GA was 63% when compared to group treated with LN which exhibited 81% efficiency. Systemic diverse effects were only observed with use of LN. 0.075 p was obtained (7.5%). Conclusions: GA efficiency was ascertained although it was lesser than that of LN. GA can be used as an adjuvant treatment in order to decrease lesions, it elicits minimal local adverse effects.

ABSTRACT

STUDY DESIGN: A retrospective study. PURPOSE: To evaluate the immunity-enhancing effect of implantation of a liquid nitrogen-treated tumor. OVERVIEW OF LITERATURE: We have developed a new technique of implanting a tumor frozen in liquid nitrogen after posterior decompression and stabilization, with the aim of enhancing antitumor immunity in order to prolong the survival period of the patient. In the current study, the immunity-enhancing effect of this new technique has been evaluated. METHODS: The subjects were 19 patients in whom we had earlier performed decompression and stabilization between April 2011 and September 2013. The 19 subjects were divided into two groups, namely a frozen autologous tumor tissue implantation group (n=15; "implantation group"), which consisted of patients, who underwent implantation with autologous tumor tissue frozen in liquid nitrogen, and a control group (n=4), which consisted of patients, who did not undergo autologous cancer transplantation. To evaluate the immunity-enhancing effect of the protocol, plasma cytokines (interferon [IFN]-gamma and interleukin [IL]-12) were analyzed before surgery and a month after surgery. RESULTS: The mean rate of increase in IFN-gamma was significantly higher in the implantation group (p=0.03). Regarding IL-12, no significant difference was observed between the groups, although the implantation group exhibited increased levels of IL-12 (p=0.22). CONCLUSIONS: Decompression and stabilization combined with autologous frozen tumor cell implantation can enhance cancer immunity in metastatic spinal tumor patients. It is hypothesized that this procedure might prevent local recurrence and prolong survival period.

ABSTRACT

Objective: To explore the application of high hydrostatic pressure in the development of melanoma vaccine.Methods:The high hydrostatic pressure,liquid nitrogen freezing and thawing were used to break the murine B16 melanoma cells and then the cell suspension was mixed with Freund's adjuvant to prepare vaccine for immunizing the mice.Results:After immu-nization,the murine B16 melanoma cells were injected intravenously and subcutaneously.The immune results of the vaccines were evaluated,by comparing survival time of the mice, subcutaneous tumor volume, DTH experiments and the fluorescence imaging of tumors.Conclusion:Compared with the method of liquid nitrogen freeze-thaw broken,high hydrostatic pressure crushing cells has more advantages in the development of tumor vaccine.

ABSTRACT

Objective To verify whether the storage bag can reach the use requirement of cord blood freezing storage by conduc-ting a series of tests before staring use of new type cryopreservation bag for cord blood.Methods According to the standard opera-tion procedure,28 new type cryopreservation bags were extracted.The cord blood units were performed the cryostorage and thaw for rewarming according to the standard operating procedures.Then the physical integrity and various quality indicators of cord blood in bag were detected,including the nucleated cell viability,cell recovery rate,CD34-positive cell,colony-forming cultivation of stem cells,sterility test and verification of actual load volume.Results All the tested storage bags passed the integrity test,and the various testing indicators of the preserved cells conformed to the use requirements.Conclusion The new type storage bag has the same performance and effect to original bag,it is verified that the maximum loading volume of 50 mL(for 50 mL specification of storage bag)and 80mL(for 250mL specification of storage bag)is safe.

ABSTRACT

Objective To evaluate the clinical efficacy and safety about carbon dioxide fractional laser combined with liquid nitrogen cryotherapy for flat warts.Methods Eighty-three patients with flat warts were divided into 2 groups according to the treatment method,42 patients in treatment group were treated by carbon dioxide fractional laser combined with liquid nitrogen cryotherapy,41 patients in control group were treated by liquid nitrogen cryotherapy alone.As a period for 10 days,2 groups were given 2 periods of treatment.The therapeutic effect,recurrence rate and adverse reaction were compared between 2 groups.Results The cure rates of 1-period and 2-period in treatment group were significantly higher than those in control group [73.81%(31/42) vs.46.34%(19/41) and 97.62%(41/42) vs.75.61%(31/41)],the recurrence rate and incidence of adverse reaction in treatment group were significantly lower than those in control group [2.44%(1/41) vs.19.35%(6/31) and 9.52%(4/42) vs.31.71%(13/41)],there were statistical differences (P < 0.05 or < 0.01).Conclusion Carbon dioxide fractional laser combined with liquid nitrogen cryotherapy for fiat warts is a safe and effective treatment with low recurrence rate.

ABSTRACT

Se realizó un estudio descriptivo y transversal de 200 pacientes con leucoplasia bucal, atendidos en la consulta estomatológica del Policlínico de Especialidades del Hospital Provincial Docente Clinicoquirúrgico "Saturnino Lora Torres" de Santiago de Cuba, desde septiembre del 2007 hasta igual mes del 2012, a fin de evaluar los efectos terapéuticos del nitrógeno líquido en el tratamiento de esta afección. Este refrigerante se empleó en la remoción quirúrgica de la citada enfermedad y propició una óptima regeneración de los tejidos bucales afectados en el menor tiempo posible. Para ello se aplicaron intervenciones repetidas de enfriamientos súbitos, así como deshielos lentos y espontáneos, con excelentes resultados en los integrantes de la casuística. Esta sustancia criogénica fue efectiva en el epitelio mucoso por sus propiedades, tales como: anestésica, antimicrobiana, antiinfecciosa, cicatrizante e inmunológica y permitió una curación rápida, menos dolorosa, sin riesgo ni complicación e inocua a los tejidos bucales, lo cual impidió la formación de cicatrices residuales.


A descriptive and cross-sectional study was carried out in 200 patients with oral leukoplakia, attended in the stomatology department of the Polyclinic of Specialties in "Saturnino Lora Torres" Provincial Teaching Clinical Surgical Hospital of Santiago de Cuba, from September 2007 to the same month of 2012, in order to evaluate the therapeutic effects of liquid nitrogen in treating this condition. This coolant was used in the surgical removal of the lesion and contributed to a good regeneration of affected oral tissues in the shortest time possible. With this purpose repeated interventions of sudden cooling were applied, as well as slow and spontaneous thaws with excellent results in the members of the case material. This cryogenic substance was effective in the mucosal epithelium by its anesthetic, antimicrobial, anti-infective, healing and immunologic properties, allowing a quick and less painful cure, without risk or complication, and safe for oral tissues, which prevented the occurrence of residual scars.

ABSTRACT

The aim of the present study was to investigate the efficiency of the encapsulation-dehydration technique for cryopreservation of Cyrtopodium hastchbachii Pabst seeds. Immature seeds of this species were cryopreserved by an encapsulation-dehydration technique. Seeds of five immature pods, 120 days after pollination, were encapsulated in 3% calcium alginate matrix and pretreated in liquid medium supplemented with 0.08 M sucrose (24 h), 0.15 M sucrose (24 h), 0.25 M sucrose (48 h), 0.5 M sucrose (24 h) and 0.75 M sucrose (24 h) in shaker at 60 rpm. Alginate beads were dehydrated 5 h in silicagel and immersed in liquid nitrogen for 12 h. Cryopreserved beads were thawed at 30°C for 1 min, rehydrated using the same liquid mediums (0.75 M sucrose (24 h), 0.5 M sucrose (24 h), 0.25 M sucrose (48 h) and 0.15 M sucrose (24 h)) and cultivated in half strength Murashige & Skoog medium (1962) with the addition of 2 g/L activated charcoal. Sixty four percent of seeds survived and developed into acclimatized plants after being cryopreserved. In this work, the encapsulation-dehydration technique was employed for first time in Cyrtopodium hatschbachii.
